Effects of sertraline treatment on saphenous vein contraction

AIM: Selective serotonin reuptake inhibitor (SSRI) group of anti-depressant drugs are commonly used in patients with cardiovascular disorders in order to diminish anxiete or depression. However, their effects on saphenous veins are not investigated particularly in vitro. We aimed to investigate the effects of sertraline in human saphena. MATERIAL AND METHODS: Human saphenous veins (n=12) were taken from patients who underwent coronary bypass surgery. The patients' ages were between 47 to 72. Saphena tissues were cleaned and sectioned into 3-4 mm long rings. Rings were placed in organ baths containing Krebs solution, thermoregulated at 37oC and aerated (95 % O2 and 5% CO2). Changes in isometric tension of saphena were recorded using a four channel force displacement transducer. All tissues washed for 3 hours in order to diminish the effects of anaesthetic agents. Phenylephrine (PE 10-6M) was administered in tissue cabs for producing isometric contractions. Contraction with measurements were used as contraction parameters. Cumulative sertraline (10-9 to 10-3) doses were added in organ baths. The contractions were recorded accordingly. Friedman and Kruskal Wallis tests were used for statistical evaluation. Amplitudes of contractions were determined and used for statistical analyses. RESULTS and CONCLUSIONS: Sertraline treatments at 10-9, 10-8, 10-7, 10-6, 10-5 doses had no significant changes on muscle contractility. But sertraline inhibited the contractions at 10-4 and 10-3 M dose following PE administration significantly (p<0.05). This effect of sertraline occured in a dose dependent manner. This means we can use SSRI group of antidepressants in these doses confidently. These findings have a potential to contribute to studies owing to effect of sertraline on cardiovascular system.
