The objective of the present study was to evaluate the efficacy of green tae polyphenol EGCG on ageing mediated mitochondrial alteration and its associated induction of apoptosis in cardiomyocyte of aged Wistar albino rat. The Wistar albino rat was divided into four like group – I: control, group – II: aged control, group III: aged animal supplemented with EGCC 300 mg/kg b.w. for 30 days, group IV: control animal administrated with 300 mg/kg b.w. of EGCG for 30 days. At the end of the 30 days animal were sacrificed and subjected to various analysis electron microscopy analysis of intercalated disc, expression of junction protein (cadherin, α catenin, Cx43) and gene was studied by immunoblotting, immunohistochemistry and real-time PCR analysis. The result of electron microscope study of that aged animal showed marked alteration in structure of intercalated disc when compared to control. Expression of junction protein (p<0.001) and gene expression (p<0.001) significantly reduced in aged animal when compared to control. However, this alteration was significantly reversed in EGCG supplemented rat when compared to aged rat. No significant alteration was observed in control animal treated with EGCG. Taken together the present data along with histological finding clearly emphasize that the EGCG potentially inhibit or reverse the ageing mediated intercalated disc remodeling thereby it protect the heart from ageing mediated cardiac hypertrophy and remodeling.
