In today’s industrialized society we are exposed to various environmental hazardous materials. One of these highly toxic metal is Cadmium (Cd), is known to affect various mammalian organs like, kidney, liver, lung, pancreas, prostate, ovaries, placenta etc, including testis1 . It induces oxidative stress which is one of the most important causes leading to male infertility. Herbal remedies like, Moringa tree has a great therapeutic value & is found to be effective both in prevention and treatment modalities, to combat toxicity of these materials 2. This study is conducted to evaluate the influence of Pre & Post treatment with Moringa oliefera leaf extract (MoE) on testicular damage in cadmium exposed male Wistar Albino rats. All procedures were performed as per the guidelines established by the Institutional Animal Ethics Committee. Wistar Albino rats weighing between (180-200) g were broadly divided into 5groups, each having six (6) animals, as follows: Group i – control group, received normal saline only. Group ii – experimental control group, pre- treated (oral) with M. oleifera leaf extract, 100 mg/kg/bw) for 10 days. Group iii – received a single oral dose of cadmium chloride (10 mg/kg/bw). Group iv – pre – treated with M. oleifera leaf extract (100 mg/kg/bw) for 10 days, followed by cadmium chloride (10 mg/kg/bw) given orally for one day. Group v: Received a single dose of cadmium chloride (10mg/kgbw) followed by Moringa olifera leaf extract (100mg/kgbw ) 10 days Using 23 G needles, 2 ml of blood was collected directly via cardiac puncture, for the estimation of the testosterone levels, under aseptic precautions, The testes were removed, homogenized and centrifuged for 10 minutes. The supernatant obtained was used for the Malondialdehyde (MDA) estimation4. After the experimental procedure they were sacrificed by administering sodium pentobarbitone, 40 mg/kg The data were expressed as means ±SD was analyzed using students T test . P<0.05 were considered as statistically significant Treatment with MOE prior & post treatment with aqueous moringa olifera leaf extract on tissue lipid peroxidation level & serum testosterone level. A significant decrease in testosterone and increase lipid peroxidation was observed (p < 0.0001) in cadmium treated group when compared to the control and pre & post treated M. oliefera leaf extract (MoE), whereas a significant decrease in testicular lipid peroxidation and increase in testosterone levels were seen when group iv & group v was compared to group iii. The plant extract thus provides a cheap alternative in treating testicular damage caused due to the exposure to cadmium and sexual dysfunction. in sex hormone and testicular damage. Therefore, the present study suggests that though both of the treatment modalities with Moringa olifera leaf extract altered the testosterone & tissue lipid peroxidation levels, pre-treatment showed better outcome in combatting cadmium toxicity.
