Single-cell fluorescence-based measurements of cytosolic free calcium ([Ca2 +]i) have now become commonplace, accounting, for instance, for roughly 15% of the papers in two recent Volumes of the Journal of Physiology. So it is easy to forget that the “single cell [Ca2+] revolution” began less than fifteen years ago with the landmark invention of Fure-2 and Indo-1 by Roger Tsien and his co-workers (Grynkiewicz et al., 1985).
